TROP2 (trophoblast cell-surface antigen 2, encoded by TACSTD2) is a transmembrane glycoprotein overexpressed across many epithelial solid tumors — breast (including triple-negative and HR-positive/HER2-negative disease), lung, and urothelial cancers among them — while its expression in normal tissue is more limited. High TROP2 expression tends to track with more aggressive disease, and its accessibility at the cell surface has made it one of the most active antibody-drug conjugate targets in oncology.
The target is clinically proven. Three TROP2 antibody-drug conjugates have reached approval — sacituzumab govitecan (Trodelvy), datopotamab deruxtecan (Datroway), and sacituzumab tirumotecan (sac-TMT) — and behind them sits a widening pipeline of next-generation ADCs, bispecific antibodies, aptamer-drug conjugates, and CAR-NK cells.
