CCR8 is a chemokine receptor that stands out for where it is expressed: it sits preferentially on regulatory T cells (Tregs) that have infiltrated the
tumor, while staying low on Tregs and other immune cells in healthy tissue and blood. Because tumor-infiltrating Tregs suppress local antitumor immunity, that restricted pattern makes CCR8 a way to reach and remove the suppressive cells inside the tumor without broadly disturbing Tregs elsewhere.
Most programs are antibodies engineered to deplete CCR8-positive Tregs through enhanced antibody-dependent cell cytotoxicity, alongside bispecific antibodies, antibody-drug conjugates, CAR-T cells, and small molecules. The field is still early — programs span preclinical work and early clinical trials — but interest has grown quickly as a way to complement checkpoint blockade in solid tumors.
